Sentynl to License Investigational Molecule from PRG S&T for Progeria Treatment

The two sides will begin immediately to advance clinical development of Progerinin for Hutchinson-Gilford Progeria Syndrome.

Sentynl Therapeutics has agreed to license PRG S&T’s investigational molecule, Progerinin (SLC-D011), for the treatment of Hutchinson-Gilford Progeria Syndrome (HGPS). The U.S. FDA has already designated Progerinin as an orphan drug.

Progeria Defined

HGPS or progeroid laminopathies, collectively called progeria, are ultra-rare and fatal genetic premature aging diseases accelerating mortality in young patients. Children who have HGPS typically die of atherosclerosis, a heart disease that affects millions of adults who age normally.

However, in these children’s cases, the average age of death is 14.5 years. Preceding this period are numerous disease manifestations. These include scleroderma-like skin, global lipodystrophy, alopecia, joint contractures, skeletal dysplasia, debilitating strokes, and an overall severe failure to thrive.

How Investigational Molecule Helps

The root cause of HGPS is accumulation of progerin. This is an abnormal form of the lamin A protein, produced by mutations in the LMNA gene. Ultimately, that abnormality disrupts nuclear structure, leading to premature cellular aging.

Progerinin, an orally active, small-molecule drug, inhibits interactions and harmful effects of progerin within cells. Its intent is to improve nuclear integrity and reduce cellular damage. Early research and clinical trials aim to determine if Progerinin can slow disease progression and improve survival among HGPS patients.

Terms of Sentynl, PRG Agreement

Under the agreement, Sentynl, a California-based biopharmaceutical company and subsidiary of Zydus Lifesciences, can begin working with PRG S&T immediately. The two sides intend to advance SLC-D011’s clinical development; the program is currently finalizing a Phase IIA clinical trial. Data are expected before the end of the first half of 2026.

If certain milestones are met, Sentynl will acquire full rights to the molecule for HGPS upon closing of the agreement. Progerinin would join Zokinvy (lonafarnib) as Sentynl’s second therapy intended for treatment of HGPS.
